چکیده

A highly malignant human T-cell leukemia was identified by cell surface analysis as a member of the T-cell receptor (TCR) yS lineage. Cytogenetic and molecular analysis showed a novel t(8;14)(q24;qll) rearrangement involving the J,, gene segment on chromosome 14 and the distal end of chromosome 8 near the c-myc proto-oncogene locus. The yS TCR of the leukemia blasts was functionally intact and could be activated to generate intracellular calcium flux and to target Fc receptor-mediated redirected tumor cell lysis. In addition,

A New Translocation , t ( 1 O ; 14 ) ( q 24 ; qll ) , in T Cell Neoplasia

Four cases of T cell neoplasia are reported: three presenting as T cell acute lymphoblastic leukemia and one presenting in the leukemic phase of a T cell lymphoma. In all cases, the cells of the leukemic clone were characterized by an identical cytogenetic abnormality. This abnormality was a unique reciprocal translocation involving chromosomes 10 and 1 4. The breakpoint in chromosome 1 4 was i...

Characterization of a novel human natural killer-cell line (NK-YS) established from natural killer cell lymphoma/leukemia associated with Epstein-Barr virus infection.

A novel cell line was established from a patient with a leukemic-state nasal angiocentric natural killer (NK) cell lymphoma with systemic skin infiltration. The morphology of the leukemic cells was large-granular-lymphocyte (LGL), and their immunophenotype was CD2+, CD3-, CD5+, CD7+, CD16-, CD56+, and CD57-. The presence of Epstein-Barr viral (EBV) genome was shown in specimens from the patient...

Antileukemia activity of a natural killer cell line against human leukemias.

We describe here the in vitro and in vivo antileukemia activity of a recently described natural killer (NK) cell line (NK-92), which has features of human activated NK cells. The cytotoxic activity of rhIL2-dependent cultured NK-92 cells against primary patient-derived leukemic target cells [12 acute myelogenous leukemias (AMLs), 7 T acute lymphoblastic leukemias (T-ALLs), 14 B-lineage-ALLs, an...

Bortezomib treatment and regulatory T-cell depletion enhance the antitumor effects of adoptively infused NK cells.

Ligation of inhibitory receptors renders natural killer (NK) cells inactive against autologous tumors. Recently, the proteasome inhibitor bortezomib was shown to sensitize tumors to autologous NK-cell cytotoxicity in vitro. Here, we show bortezomib augments the antitumor effects of syngeneic NK-cell infusions in tumor-bearing animals; this effect is further enhanced in regulatory T cell (Treg c...

Distinctive immunophenotypic features of t(8;21)(q22;q22) acute myeloblastic leukemia in children.

Thirty cases of newly diagnosed pediatric acute myeloblastic leukemia (AML) with French-American-British (FAB) M2 morphology were analyzed with cytogenetics and a comprehensive panel of monoclonal antibodies reactive with lymphoid-, natural killer (NK)-cell-, and myeloid-associated antigens. The t(8;21)(q22;q22), or t(8;21;V)(q22;q22;V), translocation was identified in 16 of the 30 cases. Cases...
